Human embryonic stem cells followed by tracking of nuclei

An overnight recording on Hoechst 33342-stained hESCs was processed with segmentation and tracking in Image Analyst MKII. The cell divisions were automatically annotated with the Special/Count Cell Division and Cell Death function in the tracked time series. Related to (14).

Calcium oscillations in tracked cells

A mammalian cell culture expressing a nuclear red fluorescent protein and a cytosolic yellow cameleon calcium sensor was overnight imaged and processed with segmentation and tracking in Image Analyst MKII. Nuclei were then colored uniformly to the measured calcium ratio using the Measure Object Intensity function.

Segmentation of human fibroblast culture

Human fibroblasts were stained with Hoechst 33342 and phalloidin-Alexa Fluor 647 and segmented using the nuclear stain as seed. Solid colors indicate different segments (cells).

Image segmentation of human dispersed pancreatic islet cell cultures

Live human dispersed pancreatic islet cell cultures were stained with Hoechst 33342, MitoSOX and MitoTracker Deep Red, and after fixation for insulin immunofluorescence. Image segmentation was performed to selectively revel MitoTracker and insulin positive cells, seeded by the nuclear stain. This was used to determine MitoSOX:MitoTracker Deep Red ratio selectively in pancreatic β-cells in (26).

Detection of subcellular protein aggregates

Protein aggregates revealed in Huntingtin-GFP expressing human cells using a combination of high pass filtering and segmentation classifiers in Image Analyst MKII.
